      If you want to practice in silico cloning, first watch the DNA Atlas tutorial video from this same page and then try to digest and ligate the constitutive promoter plasmid with the GFP cassette in DNA Atlas. Some of you have already done this in Biolab I, but you can still do this exercise if you like. If you need more challenge, you can search different plasmids from the iGEM resources, digest and ligate them and create your own novel in silico recombinant plasmids. There are also the common BioBrick primer sequences provided, the same that you are using in colony PCR.
